Isolation and chemical characterization of a cell-surface sialoglycopeptide fraction from novikoff ascites cells.

چکیده
Sialic acid has been shown to contribute significantly to the cell-surface charge of tumor cells. In order to elucidate the chemical nature of the sialic acid-containing molecules present on the tumor cell surface, a procedure for the isolation of a Sialoglycopeptide fraction from the surface of Novikoff ascites cells has been developed. Digestion of tumor cell suspensions with papain liberates a Sialoglycopeptide fraction from the cell surface. This fraction was partially purified using trichloroacetic acid precipitation, dialysis, and gel filtration on Sephadex G-50. The Sialoglycopeptide fraction contains 65—80%of the neuraminidase-labile sialic acid present on the surface of these cells. Quantitative analysis of the composition of this fraction revealed the presence of the following monosaccharides, expressed per mg glycopeptide: 0.39 /Limole sialic acid, 0.41 ¿/moleglucosamine, 0.23 ¿/molegalactosamine, 0.15 ¿/mole mannose, 0.54 jumÃ3le galactose, 0.53 ¿/mole glucose, and 0.15 ¿¿mole uronic acid. Amino acid analysis indicated a peptide content of 15%. Amino acids present in the highest amounts were threonine, aspartic acid, glutamic acid, serine, proline, alanine, and valine (molar ratio, 4:2:1.5:1:1:1:1).

Isolation and Chemical Characterization of Cell Surface Sialoglycopeptide Fractions during Progression of Rat Ascites Hepatoma AS-30Dl
In order to ascertain whether chemical alterations in cell surface glycopeptides may be associated with tumor progression, we isolated Sialoglycopeptide fractions from the surface of AS-30D rat ascites hepatoma cells before and after tumor progression. During the 70th to 90th transplantation generation of the AS-30D tumor, a morphological change and an increase in tumor virulence were observed....

متن کاملIsolation and in vitro Characterization of Mesenchymal Stem Cells Derived from the Pulp Tissue of Human Third Molar Tooth
Background: It is still controversial that the stem cells isolated from human dental pulp meets the criteria for mesenchymal stem cells (MSCs). The aim of the present study was to examine whether or not they are MSCs, or are distinct stem cells population residing in tooth pulp. Methods: Adherent fibroblastic cells in the culture of pulp tissue from human third molars were propagated through se...
متن کاملChanges in the PPla mRNA Level and PP1 Activity of Ascites Hepatomas at Different Cell Growth Rates
It is now well established that the type la of serine/threonine protein phosphatases (PP1α) may be implicated in malignant phenotype. Although the PP1α mRNA level increased in livers at preneo-plastic stages of hepatocarinogenesis and all of examined rat ascites hepatomas, unexpectedly, dra-matically decreased in some of primary hepatomas. In order to elucidate the low level of PP1α mRNA in som...
متن کاملIsolation and Characterization of Human Induced Pluripotent Stem Cells-Derived Mesenchymal Progenitors
Purpose: Isolating human induced pluripotent stem cells (hiPS)-derived mesenchymal progenitors as a new source of mesenchymal cells which can differentiate into different lineages like adipose and bone. Materials and Methods: After 7 days of hiPS1 culture on matrigle coated dishes, spindle like cells around colonies were removed by cell scraper. These cells that had mesenchymal like morphology ...
